Table 1 CD34+ cell transduction with MND constructs. (A). 1 × 103 and 2 × 10 3 transduced CD34+ cells transduced with the control or U16RBE expression vectors were plated into 1 ml semi-solid methylcelluose media. After 14 days, colonies were counted and analyzed for Erythroid-Burst Forming Units (BFU-E), Granulocyte Macrophage -Colony Forming Units (CFU-GM) and Granulocyte Erythroid Macrophage Megakaryocyte Colony Forming Units (CFU-GEMM). (B) After transduction CD34+ cells were allowed to expand in stroma flasks, and cell samples were taken for eGFP FACS analysis 1 and 45 days after plating to assess transduction frequency. The loss of eGFP expression with time is a consequence of epigenetic silencing of the MND LTR (Castanotto, Li and Rossi, unpublished results).
